Second cast does not. the first cast of "Clarity of Will" without "Grace" benefit is at 300k the second cast of "Clarity of Will" ,while the first cast still affect the target, upgrades it to 600k the first cast of "Clarity of Will" with "Grace" benefit brings it to 400k the second cast of "Clarity of Will" with "Grace" benefit, while the first cast still affects the target, upgrades it to just 600k.
